The Case for Nootropic Peptides

Traditional nootropics like racetams, modafinil, and caffeine work primarily by modulating neurotransmitter levels or receptor sensitivity. Nootropic peptides take a fundamentally different approach: they influence brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and other signaling molecules that govern neuroplasticity, neurogenesis, and neuroprotection. The result is not just temporary cognitive enhancement but potentially lasting improvements in brain structure and function.

Semax: The Focus Peptide

Semax is a synthetic analog of adrenocorticotropic hormone (ACTH 4-10) developed in Russia, where it has been approved as a prescription medication for conditions including stroke recovery and cognitive disorders. It is a seven-amino-acid peptide that crosses the blood-brain barrier when administered intranasally.

Semax's cognitive effects are mediated through several pathways: it increases BDNF expression (critical for learning and memory formation), modulates serotonin and dopamine metabolism, enhances attention and information processing, and provides neuroprotective effects against oxidative stress. Users commonly report improved focus, mental clarity, verbal fluency, and motivation within 15-30 minutes of intranasal administration.

Standard dosing is 200-600 mcg intranasally, 1-2 times daily. The NA-Semax variant (with an added acetyl group) is reported to be more potent at lower doses.

Selank: The Anti-Anxiety Nootropic

Selank is a synthetic analog of the immunomodulatory peptide tuftsin, also developed in Russia and approved for clinical use there. It acts on GABAergic pathways, modulating anxiety and stress response without the sedation or cognitive impairment associated with benzodiazepines. This makes Selank uniquely valuable for people whose cognitive performance is hindered by anxiety or stress.

Beyond anxiolysis, Selank enhances memory consolidation, increases BDNF expression, and has immunomodulatory properties. It is particularly useful for high-pressure cognitive tasks where anxiety can degrade performance. Common dosing is 200-400 mcg intranasally, 1-3 times daily.

Dihexa: The Potent Neurotrophic

Dihexa is a hexapeptide derived from angiotensin IV that has shown extraordinary potency in preclinical studies. Research from Washington State University demonstrated that Dihexa is approximately 10 million times more potent than BDNF at promoting new synaptic connections. It works by activating the hepatocyte growth factor (HGF)/c-Met receptor system, promoting dendritic spine formation, synaptogenesis, and potentially neurogenesis.

Dihexa is administered orally or sublingually, typically at very low doses (10-20 mg). However, its extreme potency and limited human safety data make it the most experimental compound in this stack. Caution is strongly warranted, and many in the peptide community consider it an advanced-only option.

Why These Three Synergize

  • Complementary BDNF pathways: Semax and Selank both increase BDNF but through different mechanisms, while Dihexa acts through the HGF/c-Met system for synaptogenesis.
  • Anxiety and focus balance: Semax enhances dopaminergic drive and focus, while Selank reduces anxiety that can impair performance. Together they create a calm-focused state.
  • Short-term and long-term effects: Semax provides immediate cognitive enhancement, while Dihexa and ongoing BDNF elevation from all three may produce lasting structural brain improvements.

Sample Cognitive Stack Protocol

  • Morning: Semax 300-600 mcg intranasal + Selank 200-400 mcg intranasal
  • Optional (advanced): Dihexa 10-20 mg oral/sublingual once daily
  • Cycle: Semax and Selank can be used for 2-4 weeks on, 1-2 weeks off. Dihexa cycles are typically shorter (2-4 weeks) given its potency.

Safety Considerations

Semax and Selank have the strongest safety profiles of the three, with decades of clinical use in Russia. Side effects are generally mild and may include nasal irritation, headache, or dizziness. Dihexa has far less human safety data and should be approached with extreme caution. Its potent effects on growth factor signaling raise theoretical concerns about long-term use. Always start with the lowest effective dose and monitor your response carefully.
